چکیده
A unique capsidless virus with a positive-sense, single-stranded RNA genome (hadakavirus 1, HadV1), member of the extended picorna-like supergroup, was isolated previously from phytopathogenic fungus Fusarium oxysporum. Here, we describe molecular and biological characterisation second hadakavirus strain nygamai, which has not been investigated in detail as host. This virus, 1 1NL (HadV1-1NL), features similar to first hadakavirus, HadV1-7n, despite having different number segments (10 for HadV1-1NL vs. 11 HadV1-7n). The 10 genomic range size 0.9 kb 2.5 kb. All show 67% 86% local nucleotide sequence identity their HadV1-7n counterparts, whereas no homolog RNA8, encodes zinc-finger motif. Another interesting feature is possible coding incapability RNA10. predicted be based on RNase susceptibility its replicative form dsRNA. Phenotypic comparison multiple virus-infected virus-free single-spore isolates indicated asymptomatic infection by HadV1-1NL. Less-efficient vertical transmission via spores observed infected fungal colonies were derived became older, HadV1-7n. study shows example that appears have unusual features.

“ISA-Lation” of Single-Stranded Positive-Sense RNA Viruses from Non-Infectious Clinical/Animal Samples
Isolation of viral pathogens from clinical and/or animal samples has traditionally relied on either cell cultures or laboratory animal model systems. However, virus viability is notoriously susceptible to adverse conditions that may include inappropriate procedures for sample collection, storage temperature, support media and transportation. Using our recently described ISA method, we have deve...
متن کاملDouble-stranded RNA mycovirus from Fusarium graminearum.
Double-stranded RNA (dsRNA) viruses in some fungi are associated with hypovirulence and have been used or proposed as biological control agents. We isolated 7.5-kb dsRNAs from 13 of 286 field strains of Fusarium graminearum isolated from maize in Korea. One of these strains, DK21, was examined in more detail. This strain had pronounced morphological changes, including reduction in mycelial grow...

A novel positive-sense, single-stranded RNA (+ssRNA) virus (Halastavi árva RNA virus, HalV; JN000306) with di-cistronic genome organization was serendipitously identified in intestinal contents of freshwater carps (Cyprinus carpio) fished by line-fishing from fishpond "Lőrinte halastó" located in Veszprém County, Hungary. The complete nucleotide (nt) sequence of the genomic RNA is 9565 nt in le...
متن کاملSingle-stranded positive-sense RNA viruses generated in days using infectious subgenomic amplicons
Reverse genetics is a key methodology for producing genetically modified RNA viruses and deciphering cellular and viral biological properties, but methods based on the preparation of plasmid-based complete viral genomes are laborious and unpredictable. Here, both wild-type and genetically modified infectious RNA viruses were generated in days using the newly described ISA (infectious-subgenomic...
متن کاملThe relationship between mutation frequency and replication strategy in positive-sense single-stranded RNA viruses.
For positive-sense single-stranded RNA virus genomes, there is a trade-off between the mutually exclusive tasks of transcription, translation and encapsidation. The replication strategy that maximizes the intracellular growth rate of the virus requires iterative genome transcription from positive to negative, and back to positive sense. However, RNA viruses experience high mutation rates, and t...
